High‑res diagram: GSDMD regulates pathways mediating OA lesions

🔍 Mechanism of this paper

GSDMD is up‑regulated in osteocytes within the subchondral bone during osteoarthritis. Under inflammatory stimulation, GSDMD‑deficient osteocytes trigger excessive activation of the CARD9/NF‑κB signalling pathway, inducing robust secretion of inflammatory mediators and osteoclastogenic factors. This disturbs the coupling balance between osteoblasts and osteoclasts, gives rise to high‑turnover subchondral bone remodelling, alters joint mechanical microenvironment, and consequently exacerbates cartilage degeneration.

💡 Innovations & Breakthroughs

This study overturns the established notion that GSDMD solely acts as an executioner of pyroptosis. Distinct from global gene‑knockout models, osteocyte‑specific knockout mice are adopted to uncover the bone‑homeostasis‑regulating function of GSDMD in osteocytes. It confirms that chondrocytes can activate pyroptosis‑related signalling in osteocytes. The regulatory effects of osteocyte‑derived GSDMD on bone remodelling are verified in both osteoarthritis and osteoporosis animal models.

🧠 In‑depth Insights

The same molecule can exert functionally heterogeneous effects across different cell types. Reduced osteocyte death does not equal ameliorated osteoarthritis pathology. Aberrant subchondral bone remodelling acts as a critical driving force for OA progression.
