Epoxy-Sepharose FF is an activation chromatography separation medium intermediate formed by bonding epoxy groups on agarose microspheres. The product retains the excellent hydrophilicity of agarose and the large network structure, has good compatibility with biologically active macromolecules, and has high activated group density. Instructions:


¡¡ 1. Before ligand coupling, prepare all samples, solutions, buffers, etc., to minimize the gap between each step.


¡¡¡¡ 2. Wash more than 10 volumes with distilled water before coupling, and remove the preservation solution.


¡¡¡¡3. Choose the coupling solution system according to the target ligand. Generally, the pH for sulfhydryl coupling is 7-9, the pH for amino coupling is 9-11, and the pH for hydroxyl coupling is >11.


¡¡¡¡4. It can be coupled at 20-40¡ãC. Generally, increasing the temperature can accelerate the coupling. In the case of poor coupling effect, the temperature can be increased to 35¡ãC or more, but the stability of the ligand must be considered.


¡¡¡¡ 5. The coupling time depends on the reaction speed, usually it can be coupled overnight (16 h).


6. After coupling the ligand, the remaining epoxy groups need to be blocked. Use cysteine, ethanolamine, glycine and other small molecule reagents to block for several hours, such as 0.2 M Na2CO3, 0.1 M ethanolamine, pH 10, 37¡æ for 16 h .


¡¡¡¡7. After coupling, wash with pH 8-9, 0.1 mol/LTris-HCl buffer and pH 4-5, 0.1 M acetic acid-sodium acetate buffer in turn, and cycle three times to remove uncoupled residual ligands.


¡¡¡¡ 8. The coupled gel can be stored in a solution containing a bacteriostatic agent, such as 20% ethanol.
